XYZ-difference
This function is available only for fluorescence settings. It can define focus differences
among filters to achieve more precise focusing. The XYZ difference feature finds the
appropriate XYZ positions when the signal of the different fluorophores colocalize.
The refraction of the lens depends on the wavelength of the transmitted light, and it
results in different focus points in XYZ for different fluorophores. To adjust the different
focus points, it is recommended to use a slide which is loaded with fluorescent beads (e.g.
Invitrogen FocalCheck, F36909), so the XYZ position of the beads can easily be defined in
each fluorescent channel. To this purpose, find the focus pane for the beads in each
channel and then click
Store X/Y-position
in addition to the
Z-position
.

Mark the round checkbox in the
Reference
column to select a filter. It is only
possible to select a reference filter after finding the related XYZ position.

Click
Grab reference image
to grab/save the image belonging to the reference
filter.
•
Mark the
Show reference image
checkbox to overlap each live image of the
corresponding filter with the reference image to be able to compare them.
After defining the X/Y/Z-positions and calculating the difference values automatically, click
Save
.

Image compensation
You can use a saved compensation image whenever compensation-related issues occur
(deriving from the slide, illumination, staining or embedding medium irregularities).
