Use these tools to align and calibrate a laser set (laser, filter, and grating) or to create or edit the
video calibrations for your microscope objectives.
Aligning and Calibrating Laser Sets
Use the Align/Calibrate feature in the OMNICxi software to align the installed laser, filter
and grating and to calibrate the laser wavelength. If you have multiple lasers, gratings and
filters, each laser, grating, and filter set must have associated alignment and calibration data.
Once the alignment and calibration data are created, you can interchange your laser sets
without recalibrating them.
Recalibrate all of your laser sets in the following situations:
• It has been more than one week since the laser sets were aligned and calibrated. We
recommend aligning and calibrating once a week.
• You have just moved the instrument.
• You see an error message related to the detector signal intensity (no signal or signal too
low) or spectral quality (peaks too broad, peaks in wrong position, artifacts in the
spectrum), your spectral search match numbers are lower than expected or the
performance check failed.
To ensure the accuracy of your data, OMNICxi software performs automatic x-axis
calibration every 30 minutes to account for any changes in the temperature of the
environment around your instrument.
A notification alerts you when the x axis is being calibrated, and calibration typically takes
about 1 minute to complete. When calibration is complete, the software resumes any task that
was interrupted.
You can turn automatic calibration on or off in the Instrument Settings view.
