LSM + ConfoCor
The
LSM + ConfoCor
function (Fig. 134) can be
used to enter offset values between LSM scanners
and the X,Y-stage, if the latter is used for FCS
measurements. The offset can be determined by
bleaching holes into a dye layer and determining
the offset between the defined cross lines and
actual bleach spots. Type in the corresponding
numbers in the
Offset X
,
Offset Y
and
Offset Z
dialogue boxes or use the arrowheads.
If the scanners are used, offsets due to
hysteresis can be corrected with the
Bidirectional
sliders. Numbers will only
apply to the XY-stage.
No z offset exists in the ConfoCor 3 and
hence the number in Offset Z should be 0. Z
offset is also mechanically corrected in the
factory for the ConfoCor 2. So as a rule no z
offset correction is needed for the
ConfoCor 2 and the
Offset Z
should be set
to 0.
The
Help
text explains, when positive or
negative values must be entered in
dependence where the bleach spot was
observed in relation to the cross line for a
ConfoCor system at the side port. Signs will
change for a ConfoCor system on the base
port (only applicable to ConfoCor 2).
